Rentschler Biotechnologie GmbH was once again actively involved in the Recovery conference. In this high-class, biennial meeting of specialists, current trends in the purification of biopharmaceuticals and -diagnostics are discussed. The XVII. Conference was held from June 19th to 24th, 2016 in Southampton, Bermuda.
With the topic "Smart bioprocessing" the organizers focused the attention on the close linkage of all bioprocessing steps. To understand the origin of impurities, to control them and to take into account economic terms were important issues.
Rentschler's internationally renowned downstream specialist Stefan R. Schmidt, Vice President Process Science & Production, integrated this topic directly into the title of his presentation "Modular approaches for various molecules: reinventing smart bioprocessing". Approximately 270 conference attendees followed the interesting lecture. The key issue was Rentschler’s development of a modular toolbox for the process design of the purification of complex proteins. This pragmatic approach was met with great interest.
One of the principles of the modular approach is selecting a suitable capture step with appropriate capacity shown in a case study for an Fc-fusion protein. An affinity chromatography step was combined with the use of a generic buffer system. By consequence, the preparation of eight buffer variants could be saved. Alternatively, a capture step for a highly glycosylated fusion protein using anion exchange chromatography was presented. This step provides convincing results with full maintenance of the glycosylation. Further case studies demonstrated methods for the early host cell protein reduction by precipitation and alternatives of virus inactivation for sensitive proteins.
